What Cagrilintide is

Long-acting amylin analogue

Cagrilintide is a lipidated analogue of amylin, the pancreatic hormone co-secreted with insulin that signals fullness to the brainstem. Novo Nordisk developed it as a once-weekly compound and is studying it alone and in fixed combination with semaglutide (CagriSema). It gives researchers a way to study the amylin pathway separately from the incretin pathway.

How it works

Amylin acts on calcitonin-receptor complexes with RAMP proteins, slowing gastric emptying and reducing food intake through the area postrema. Native amylin aggregates and has a half-life of minutes; cagrilintide's sequence changes prevent fibril formation and a C20 fatty-diacid chain binds albumin, extending activity to about a week.

What the research reports

A Phase 2 dose-ranging trial (Lau et al., Lancet 2021) reported mean weight reductions of up to 10.8% at 26 weeks with cagrilintide alone. Phase 3 REDEFINE trials of the CagriSema combination reported larger reductions than semaglutide alone. Preclinical work examines lean-mass preservation and the interaction with GLP-1 signalling.

Is Cagrilintide legal in Australia?

Cagrilintide is not registered by the Therapeutic Goods Administration (TGA) as a medicine in Australia and is not approved for human or veterinary use. Peptide Aus supplies it as a research reagent for in-vitro laboratory work only. Supply of any substance for human use in Australia is governed by the Poisons Standard and the Therapeutic Goods Act, and the buyer is responsible for using the material lawfully. If you are looking for a treatment, speak to a doctor.

Frequently asked questions

What is Cagrilintide?

Long-acting amylin analogue.

What is Cagrilintide studied for?

Satiety signalling: amylin-receptor pharmacology in the area postrema. Weight regulation: dose-ranging Phase 2 endpoints. Combination research: additive effects with GLP-1 agonists in REDEFINE. Gastric emptying: scintigraphy endpoints in mechanistic studies.

Cagrilintide before and after: what do the studies show?

The Phase 2 dose-ranging trial reported mean weight loss of 6.0% to 10.8% at 26 weeks across doses versus 3.0% on placebo, and the CagriSema combination reported greater reductions than semaglutide alone in REDEFINE-1.

What side effects has Cagrilintide research reported?

Gastrointestinal events, mainly nausea and vomiting, were the most common in the Phase 2 trial, with injection-site reactions also reported; most were mild to moderate.

What is the Cagrilintide dosage?

Published Cagrilintide studies specify their own amounts and routes, and those vary by model and endpoint. Peptide Aus products are supplied for in-vitro laboratory research, so we do not provide dosing, cycle or usage guidance for people. For laboratory preparation, the reconstitution calculator converts vial size and water volume into concentration and syringe units.

Cagrilintide vs retatrutide: what is the difference?

Different pathway: amylin-receptor agonism versus triple incretin/glucagon agonism. Often studied together to separate satiety signalling from incretin effects.

Cagrilintide vs tirzepatide: what is the difference?

Tirzepatide is a dual incretin agonist; cagrilintide is an amylin analogue, and the two are studied as complementary rather than competing mechanisms.

Why study cagrilintide alongside a GLP-1 compound?

The two pathways reduce food intake through different receptors, and the combination trials reported greater weight reduction than either agent alone. Research designs often compare cagrilintide, a GLP-1 agonist and the pair to separate the contributions.

5mg or 10mg?

The 5mg vial is the entry format for assay development; the 10mg vial halves the price per milligram for longer protocols.

How do you reconstitute Cagrilintide?

With bacteriostatic water added slowly down the inside wall of the vial, then swirled gently. Each vial size lists its suggested volume, and the calculator converts volume to concentration.

How should Cagrilintide be stored?

Lyophilised: −20 °C, sealed, away from light and moisture. Reconstituted: 2–8 °C, used within 8–12 weeks, never frozen.
