SLU-PP-332 is a synthetic small molecule that activates all three oestrogen-related receptors (ERRα, β and γ), nuclear receptors that coordinate the gene programs muscle switches on during endurance exercise. Developed at Saint Louis University, it is studied as an 'exercise mimetic' in preclinical models.

How it works

ERR activation increases transcription of genes for mitochondrial biogenesis, fatty-acid oxidation and oxidative muscle fibre type. In mice, the compound increased running endurance and shifted muscle toward type IIa fibres without training.

Billon et al. (ACS Pharmacol Transl Sci 2023) reported improved endurance and mitochondrial gene expression; a follow-up in diet-induced obese mice reported reduced fat gain with increased energy expenditure. All published data are preclinical.
