Preparing the kit

  1. Reconstitute MOTS-C (10mg). Add 2 mL of bacteriostatic water slowly down the inside wall of the vial and swirl gently until clear; do not shake. 10mg in 2 mL gives 5 mg/mL. The calculator has this vial preset.
  2. Reconstitute 5-Amino-1MQ (50mg). Add 2 mL of bacteriostatic water slowly down the inside wall of the vial and swirl gently until clear; do not shake. 50mg in 2 mL gives 25 mg/mL. The calculator has this vial preset.
  3. Reconstitute SLU-PP-332 (5mg). Add 1 mL of bacteriostatic water slowly down the inside wall of the vial and swirl gently until clear; do not shake. 5mg in 1 mL gives 5 mg/mL. The calculator has this vial preset.
  4. Draw and swab. Swab the vial stopper before each draw. Use a fresh U-100 syringe each time; the calculator converts the amount in milligrams to syringe units at the concentration prepared.
  5. Store. Lyophilised vials at −20 °C until reconstitution. Reconstituted solution and pens at 2–8 °C, never frozen; reconstituted vials are used within 8–12 weeks, opened bacteriostatic water within 28 days.

MOTS-C

Mitochondrial-derived peptide encoded in the 12S rRNA gene

MOTS-c is a 16-amino-acid peptide encoded inside the mitochondrial genome rather than the nuclear genome, one of a small family of mitochondrial-derived peptides discovered in the 2010s. It acts as a signal from mitochondria to the rest of the cell and is studied for metabolic regulation and exercise physiology.

Lee et al. (Cell Metab 2015) reported that MOTS-c prevented diet-induced obesity and insulin resistance in mice. Reynolds et al. (Nat Commun 2021) showed it improved physical performance in young, middle-aged and old mice. Human data are observational, showing exercise-induced increases.

5-Amino-1MQ

Small-molecule NNMT inhibitor

5-Amino-1MQ is not a peptide. It is a small quinolinium molecule that inhibits nicotinamide N-methyltransferase (NNMT), an enzyme that consumes nicotinamide and S-adenosylmethionine. Because it is supplied as a powder for reconstitution and studied alongside metabolic peptides, it sits in the same catalogue.

Rodent studies (Neelakantan et al., Biochem Pharmacol 2018) reported reduced adipocyte size, lower body-fat mass and improved metabolic markers in diet-induced obese mice without a change in food intake. Later work examined muscle stem-cell function and NAD+ levels in aged tissue.

SLU-PP-332

Pan-ERR (oestrogen-related receptor) agonist

SLU-PP-332 is a synthetic small molecule that activates all three oestrogen-related receptors (ERRα, β and γ), nuclear receptors that coordinate the gene programs muscle switches on during endurance exercise. Developed at Saint Louis University, it is studied as an 'exercise mimetic' in preclinical models.

Billon et al. (ACS Pharmacol Transl Sci 2023) reported improved endurance and mitochondrial gene expression; a follow-up in diet-induced obese mice reported reduced fat gain with increased energy expenditure. All published data are preclinical.
