Preparing the kit

1. Reconstitute CJC-1295 with DAC (10mg). Add 2 mL of bacteriostatic water slowly down the inside wall of the vial and swirl gently until clear; do not shake. 10mg in 2 mL gives 5 mg/mL. The calculator has this vial preset.
2. Reconstitute Ipamorelin (10mg). Add 2 mL of bacteriostatic water slowly down the inside wall of the vial and swirl gently until clear; do not shake. 10mg in 2 mL gives 5 mg/mL. The calculator has this vial preset.
3. Draw and swab. Swab the vial stopper before each draw. Use a fresh U-100 syringe each time; the calculator converts the amount in milligrams to syringe units at the concentration prepared.
4. Store. Lyophilised vials at −20 °C until reconstitution. Reconstituted solution and pens at 2–8 °C, never frozen; reconstituted vials are used within 8–12 weeks, opened bacteriostatic water within 28 days.

Long-acting GHRH analogue with drug-affinity complex

CJC-1295 with DAC is the same GHRH (1–29) analogue with a maleimidopropionic acid group that binds covalently to serum albumin. That extends the half-life from minutes to about six to eight days and produces a sustained elevation of GH and IGF-1 rather than discrete pulses.

Teichman et al. (JCEM 2006) reported sustained GH and IGF-1 increases in healthy adults after single and repeated doses. Development by ConjuChem for HIV lipodystrophy was discontinued. It remains a research tool for sustained GH-axis stimulation.

Ipamorelin

Selective ghrelin-receptor (GHS-R1a) agonist pentapeptide

Ipamorelin is a five-amino-acid growth-hormone secretagogue that activates the ghrelin receptor. Among the GHRP class it is notable for selectivity: in the original characterisation it released growth hormone without the cortisol, prolactin or ACTH release seen with GHRP-6 and GHRP-2.

Raun et al. (Eur J Endocrinol 1998) described the selectivity profile in rats and pigs. A Phase 2 trial of ipamorelin for post-operative ileus was completed but not pursued. Preclinical work examines bone density and gastric motility.
